Prodynorphin (or proenkephalin B) is a brain-enriched polypeptide hormone with predominant RNA and protein distribution in regions of the basal ganglia and the hypothalamus. Within the mouse CNS, moderate to strong intensity immunostaining of dense axonal networks is noticed in multiple but distinct brain regions. Strong intensity dense axonal staining is observed in the nucleus accumbens, islands of Calleja, ventral pallidum, globus pallidus, hippocampal mossy fibers, substantia nigra and the parabrachial region in the pons. Moderate-intensity axonal staining is observed in the olfactory bulb, caudate putamen, lateral septum, BNST, anterior and tuberal hypotahalamus, hypothalamic paraventricular and periventricular nuclei, ventral tegmantal area, periaqueductal gray matter and locus coeruleus region. In addition, moderate-intensity immunostaining of neuronal cell bodies is noticed in subsets of neurons in the caudate putamen, lateral septum and in the olfactory bulb mitral cells while median eminence and area postrema are strongly immunoreactive too.
